Behavior and Neuroimaging Core![]() | Training and Technical Assistance - Support | Clinical Research Resources, Behavior and Neuroimaging The mission of the Behavior and Neuroimaging Core (BNC) is to provide a coherent set of resources that will facilitate the implementation of each of the COBRE Project Leader’s research aims. The BNC provides ongoing support, training, assistance and advice to COBRE project leaders in the practical aspects of data collection, data management and data processing across a wide variety of neuroscience related methodologies. The BNC also serves as a resource to the broader neuroscience community at Brown and its affiliated hospitals and the broader Rhode Island academic community by providing formal and informal training, accessible data processing pipelines, and documentation of these and related resources. The support provided by the BNC is distinct from but complementary to services provided by the Design and Analysis Core (DAC). Whereas the DAC is focused primarily on study design, analytical modeling, and techniques for inference, the focus of the BNC is on practical aspects of research conduct such as efficient and high-quality data acquisition, implementation and deployment of standard and newly developed tools for analysis of large neuroimaging datasets, and lowering barriers to the use of such tools through direct assistance, training and documentation. Because the missions of the BNC and DAC are highly complementary, the effectiveness of each core is strengthened by collaborative relationships between them. The analytical techniques developed by the DAC in partnership with the Project Leaders are predicated upon robust high-quality data. Similarly, realization of study design principles requires practical expertise with the tools necessary to implement those principles. The BNC and DAC will share a full-time staff member with expertise in research design, statistical modeling, data management and high-performance computing. |

Drosophila Media Facility![]() | Drosophila Media Preparation | Service The Drosophila Media Facility is dedicated to providing investigators with high quality media for Drosophila research. In the centralized kitchen, a standard cornmeal, sucrose, yeast medium is prepared and dispensed into different sized culture vessels. Media is prepared according to a strict protocol that ensures consistency in diet and the absence of any bacterial, fungal or insect contamination. Researchers using glass vials return them to the facility where they are autoclaved, washed, and filled again. This centralized kitchen is equipped with a food-service quality Cleveland Range Kettle MKET-12-T. Media is pumped into trays of vials or plastic bottles using a Gilson Fly Food Dispenser. Used glass vials are returned to the facility where they are sterilized in a Steris autoclave and washed in a Northwestern Systems G403 electric heat glassware washer. |

Electron Microprobe Facility![]() | Analyzer - Electron Microprobe Contact: Stephen Parman | Cameca SX100 Electron Microprobe Analyzer The Cameca SX-100 electron microprobe is equipped with 5 wavelength dispersive spectrometers (WDS) and a Rontec energy dispersive spectrometer (EDS).The WDS spectrometers each contain two or four diffracting crystals and presently include LTAP, TAP (2), LPET (2), PET, LLIF (2), LIF, LPCO, PC1 and PC2. The last three crystals are used to measure the light elements (boron to sodium). The absolute detection limits for the instrument are approximately 50-200 ppm, depending on the element. The EDS spectrometer is capable of detecting all elements above sodium (Na). X-ray element maps can easily be acquired on the microprobe. Using 512x512 pixel maps, up to nine element maps (5 elements on WDS, 4 with EDS), plus a backscattered electron (BSE) image, can be obtained simultaneously. Maximum resolution on any one map is one micron per pixel. Utilizing the coordinate system, maps of entire thin sections can be generated. |
Electron Microscopy Facility![]() | Microscope - Focused Ion Beam Electron (FIB) | Thermo/FEI Helios Dual Beam Focused Ion Beam Electron Microscope (FIB) with an Omniprobe nanomanipulator and autoTEM sample preparation The FIB is fully equipped for (near) simultaneous ion/electron beam imaging and patterning of specimens. A Pt metal deposition source is available for ion (or electron) beam assisted deposition of conducting lines. A second source is available to enhance the carbon removal rate. An Omniprobe AutoProbe 200.2 allows the physical manipulation of samples with nanometer precision and is used along with Pt metal deposition and standard FIB trenching to allow the selection and removal of electron transparent TEM samples from specific microstructural features. A NPGS pattern generation system (plus beam blanker and external scan interface) controls the electron beam for high resolution, interlaced pattern generation used in the FIB fabrication of nanostructures. Three dimensional crystallographic data can be generated from specimens using the fully automated EBSD system (Oxford HKL&EBS-3). The microscope is also equipped with an EDS detector for elemental analysis. An electron flood gun can be used with the ion beam to image/etch nonconductive samples. A low temperature holder is available for beam sensitive samples. |
Electron Microscopy Facility![]() | Microscope - Scanning Electron (SEM) | LEO 1530 High Resolution Scanning Electron Microscope (HRSEM) The LEO 1530 VP ultra-high resolution field emitter SEM that allows for a resolution of 1 nm at a voltage of 20 kV in high vacuum and 2 nm at 30 kV in the variable pressure mode. It is fully equipped with EDS for chemical mapping and includes a Debbem tensile test stage 5kn. The instrument is equipped with Nabity pattern generating software a high speed beam blanker and is set-up for high resolution electron-beam lithography. The LEO 1530 VP is capable of a resolution of 2.1 nm at very low electron energies (1 kV) which is of great use for beam-sensitive materials like polymers and insulating surfaces. Also very useful for insulating samples is the variable operating pressure mode which allows insulating materials to be imaged without coating the surface. |
Electron Microscopy Facility![]() | Microscope - Transmission Electron (TEM) | FEI CM20 TEM The FEI CM20 is available for applications that require high tilt, moderate resolution (0.23 nm). This 200 kV analytical TEM allows the large specimen tilts needed for dislocation characterization and weak-beam imaging applications. It is equipped with a low background double tilt holder and a liquid nitrogen temperature holder. Compositional analysis is performed on this instrument using energy dispersive x-ray spectroscopy. Images are recorded using a large field of view, side mounted, digital image acquisition system. |
Electron Microscopy Facility![]() | Microscope - Scanning Transmission Electron Microscope -S(TEM) | JEOL 2100F - S (TEM) The 2100F is a 200 kV, field emission source, high-resolution/analytical S(TEM) capable of 0.10 nm lattice resolution. It is primarily used for obtaining lattice images or when chemical or crystallographic information is required from laterally small areas (1-10 nm). The instrument is equipped with an x-ray spectrometer capable of light element detection, a Gatan image filter (GIF) for element and phase analyses and enegy filtered imaging, a high tilt holder for electron tomography and a high-resolution CCD camera system for digital image acquisition and processing. A low background double-tilt holder for chemical analysis by EDS, a standard double tilt holder, a heating holder capable of 1000C, and a heating straining holder are available. Probes as small as 1 nm in diameter are accessible for micro-diffraction experiments. STEM mode for EDS line and area scans, bright and dark field STEM detectors, and high angle annular dark field is capable in STEM mode. |

There is a transfer holder so samples can be transferred to the XPS with exposure to atmosphere. |
Electron Microscopy Facility![]() | Microscope - Quattro S-E(SEM) | Thermo/FEI Quattro S-E(SEM) The Quattro is an environmental SEM which means that the sample chamber can be set to a (relatively) high pressure for water containing samples. This SEM has EDS (X-ray detection) and EBSD (electron backscatter). The Quattro S is a field emission scanning electron microscope capable of generating and collecting all available information from any type of sample material. It provides the most versatile high-resolution low-vacuum FEG SEM with extended low-vacuum capabilities and the most versatile high resolution SEM for characterization, prototyping and in situ analysis. The features include three vacuum modes enabling investigation of conductive, non-conductive and high-vacuum incompatible materials such as High-vacuum mode (<6*10-4 Pa) for imaging and microanalysis of conductive and/or conventionally prepared specimens; Low-vacuum mode (10 to 130 Pa) for imaging and microanalysis of non-conductive specimens without preparation; and ESEM™ mode (10 to 4000 Pa) for high-vacuum incompatible specimens which are impossible to investigate with traditional EM methods. The system is equipped with a 5-axes motorized x-y-z-tilt-rotate stage,for Sample navigation. |

During planning, the core can provide high quality project/trial design, including final sculpting of specific aims and hypotheses, measure selection, power analysis, data analysis planning, and grant/proposal preparation. At the beginning of the project, the core can assist in construction of REDCap data collection tools, databases, and data management, including national archiving. We provide help navigating approvals for special technology needs, such as purchasing non-standard equipment and logistics. Throughout the trial, the core can provide decision support, assessing the impact of potential design additions or changes on final statistical inference. It can serve on or help find individuals to serve on data and safety monitoring boards (DSMB). At the conclusion of the trial, the core can assist with implementation of the data analysis plan, either by providing access to contemporary statistical analysis software and coaching, or by conducting analyses for or with investigators. During reporting/publication preparation, the core can assist in write-up, data visualization, and integration of interpretations in the broader context of the project, and help address comments from peer reviewers. |

Changes in the intrinsic fluorescence of the sample are measured as a thermal ramp is applied. Changes in the fluorescence signal indicate transitions in the folding state of the protein. The Tycho can be used to study the stability of protein samples in different buffers or at different pH values. The measurement is completed in three minutes and uses a capillary tube to hold the sample. |
